pTarget(pEffector)
Expresses a guide RNA sequence to locate the desired sequence for gene knock-in. Successful in integrating ARO10 and UGT85A1 gene into the DH5α chassis genome.
pEffector is a constitutive expression transposon element assembled from pTnsABC (containing DNA fragments of TnsA, TnsB, and TnsC, please refer to TnsABC, BBa_K4761054) and pQCascade (containing DNA fragments of TniQ, Cas8, Cas7, and Cas6, please refer to TnsQ Cas876, BBa_K4761053). Furthermore, an sgRNA with a T10 insertion site is inserted to construct a transposon-targeting plasmid, which functions as a targeting element in the CRISPR RNA-guided integrase system [1].
